The development of new anti-cancer drugs that target and inhibit TrxR1 could prove to be a valuable chemotherapy tool. We hypothesized that rational-designed molecules containing a cyclic 1,2-disulfide unit as a pharmacophoric group will selectively block TrxR1 activity and will inhibit tumor growth in breast cancer.
This work describes the synthesis of two novel classes of compounds that interfere with the activity of the redox-enzyme thioredoxin reductase and were shown to inhibit in vitro breast cancer cell growth. Breast cancer is the most commonly diagnosed form of cancer in women 40-55 years of age, and it is the second major cause of cancer deaths for women in the United States.
